A handheld NASBA analyzer for the field detection and quantification of Karenia brevis

چکیده
Blooms of Karenia brevis, the red tide forming dinoflagellate in the Gulf of Mexico, cause a myriad of ecological and economic problems for coastal communities, including massive fish and mammal mortalities, and damage to tourism and fisheries/shellfish harvesting industries. There is a need for accurate detection and prediction of K. brevis blooms, including rapid and inexpensive monitoring of both water and shellfish meats to ensure the safety of shellfish harvested for human consumption. To address this issue, we have developed a protocol for easy field extraction of cellular RNA from water samples and coupled it with a handheld nucleic acid sequence-based amplification (NASBA) sensor that amplifies and detects target mRNA specific to the rbcL gene of K. brevis. This extraction protocol is a modified version of the Qiagen RNeasy Mini Kit spin protocol and requires no specialized equipment or training. Once extracted, the RNA is amplified and detected by NASBA in an in-house designed and produced handheld sensor that provides a real-time fluorescence plotting of the amplification. Both the field RNA extraction protocol and the handheld NASBA analyzer compared favorably to laboratory-based technologies. In duplicate reactions, the amplification curves generated with the handheld detector closely mirrored the curves generated with the bench top Nuclisens EasyQ NASBA analyzer and there was no difference in the sensitivity obtained using the handheld device versus the bench top models. This extraction protocol and detection sensor will be a valuable tool for rapidly monitoring K. brevis in field environments. # 2006 Elsevier B.V. All rights reserved.

Extraction and analysis of lipophilic brevetoxins from the red tide dinoflagellate Karenia brevis.
Efficient extraction and accurate analysis of lipophilic brevetoxins (PbTxs), produced by the harmful algal bloom (HAB) species Karenia brevis, are essential when assessing the toxicological potential of this dinoflagellate. One of the most commonly used brevetoxin extraction methodologies employs C18 solid-phase extraction (SPE). In this study, C18 SPEC discs were tested for extraction of spik...

Specific objectives of this research include the following: 1) Measurements of inherent and apparent optical properties will be conducted in areas frequented by Karenia brevis bloom events off the west Florida coast, and attempt to identify key optical indices for discriminating blooms of K. brevis from other phytoplankton and other sources of optical variability. 2) We will examine the utility...

Viral myocarditis is a moderate disease, but it sometimes causes progressive cardiac disorder. Many different viruses have been considered as the agent of viral myocarditis, but Coxsackievirus of the B group, in particular of the Coxsackievirus B3 (CVB3), is more than fifty percent of cases of viral myocarditis. CVB3 is a positive single-stranded RNA virus and a member of the genus Enterovirus ...

Enteroviruses are the causative agents of a number of diseases in humans. Group B coxsackieviruses are believed to be the most common viral agents responsible for human heart disease. Genomic data of enteroviruses has allowed developing new molecular approaches such as Nucleic Acid Sequence Based Amplification (NASBA) for detection of such viruses. In this study, coxsackievirus B3 (CVB3) was de...
متن کاملProgress in developing a new detection method for the harmful algal bloom species, Karenia brevis, through multiwavelength spectroscopy
Multiwavelength spectroscopy is a rapid analytical technique that can be applied to detect, identify, and quantify microorganisms such as Karenia brevis, the species known for frequent red-tide blooms in Florida’s coastal waters. This research will report on a model-based interpretation of UV–vis spectra of K. brevis. The spectroscopy models are based on light scattering and absorption theories...
